چکیده

Biological N2 fixation, the reduction of N2 to NH 3 , is catalyzed by the enzyme nitrogenase . Fractionation studies of nitrogenases from a variety of organisms have shown that all are composed of two similar proteins, one contains Mo and Fe and is called the Mo-Fe protein and the other contains only Fe and is called the Fe protein . Recently homogeneous preparations of these proteins from Azotobacter vinelandii, Clostridium pasteurianum, and Klebsiella pneumoniae have been

Interactions of heterologous nitrogenase components that generate catalytically inactive complexes.

A unique method is described for inhibiting nitrogenase. When Clostridium pasteurianum nitrogenase is assayed in the presence of the Mo-Fe protein of Azotobacter vinelandii, all the characteristic activities of nitrogenase are inhibited. C. pasteurianum nitrogenase is unaffected by the Fe protein of A. vinelandii. The Fe protein, but not the Mo-Fe protein of C. pasteurianum, inhibits A. vinelan...

Characterization of an oxygen-stable nitrogenase complex isolated from Azotobacter chroococcum.

In crude cell-free extracts of Azotobacter chroococcum, nitrogenase was much less sensitive to irreversible inactivation by O2 than was the purified enzyme. When nitrogenase was partially purified by anaerobic discontinuous sucrose-density-gradient centrifugation, O2-tolerance was retained. This preparation was considerably enriched in four polypeptides, three of which were derived from the Mo-...

The molybdenum--iron protein of Klebsiella pneumoniae nitrogenase. Evidence for non-identical subunits from peptide 'mapping'.

The molybdenum- and iron-containing protein components of nitrogenase purified from Klebsiella pneumoniae, Azotobacter vinelandii, Azotobacter chroococcum and Rhizobium japonicum bacteroids all gave either one or two protein-staining bands after sodium dodecyl sulphate/polyacrylamide-gel electrophoresis, depending on the commercial brand of sodium dodecyl sulphate used. The single band obtained...

Iron EXAFS of Azotobacter vinelandii Nitrogenase Mo-Fe and V-Fe Proteins

The structure of the iron sites of nitrogenase in dithionite-reduced and thionine-oxidized forms of the Mo-Fe and V-Fe proteins has been investigated using Fe K-edge X-ray absorption spectroscopy. For the dithionite-reduced Azotobacter uinelandii Mo-Fe protein, the dominant EXAFS Fourier transform peaks are assigned to F e 4 and Fe-Fe interactions at -2.32 and 2.64 A, as expected for F e S clus...

The Fe-V Cofactor of Vanadium Nitrogenase Contains an Interstitial Carbon Atom.

The first direct evidence is provided for the presence of an interstitial carbide in the Fe-V cofactor of Azotobacter vinelandii vanadium nitrogenase. As for our identification of the central carbide in the Fe-Mo cofactor, we employed Fe Kβ valence-to-core X-ray emission spectroscopy and density functional theory calculations, and herein report the highly similar spectra of both variants of the...
